چکیده

Abstract Objectives This study aims to investigate whether apoptosis repressor with caspase recruitment domain (ARC) could promote survival and enhance osteogenic differentiation of bone marrow-derived mesenchymal stem cells (BMSCs). Materials methods The lentivirus transfection method was used establish ARC-overexpressing BMSCs. CCK-8 detect cell proliferation. BD Pharmingen™ APC Annexin V Apoptosis Detection kit apoptosis. capacity investigated by OCN immunofluorescence staining, ALP analysis, ARS assays, RT-PCR analysis. Cells were seeded into calcium phosphate cement (CPC) scaffolds then inserted subcutaneously nude mice the defect area rat calvarium. Histological analysis conducted evaluate in vivo new formation RNA-seq possible mechanism effect ARC on Results promoted BMSC proliferation inhibited enhanced vitro. An revealed that can inhibit increase formation. regulates BMSCs mainly activating Fgf-2/PI3K/Akt pathway. Conclusions present suggests is a powerful agent for promoting regeneration provides promising tissue engineering.

Secreted Frizzled-Related Protein Promotes Bone Regeneration by Human Bone Marrow-Derived Mesenchymal Stem Cells

Secreted frizzled-related protein (sFRP)-3 is a negative regulator of Wnt signaling in human mesenchymal stem cells (hMSCs). The present study investigated the effects sFRP-3 on osteogenic differentiation by assessing osteogenic gene expression in hMSCs in vitro and by examining bone regeneration in a rat bone defect model. sFRP-3 treatment induced osteogenic differentiation in hMSCs as determi...

Study of Chondrogenic Effects of Chondrocytes Cocultured With Murine Bone Marrow-Derived Mesenchymal Stem Cells

Purpose: Co-culture systems of marrow derived mesenchymal stem cells (mMSCs) with mature chondrocytes have theoretically been considered as a putative way of MSCs chondrogenic differentiation. MSCs differentiated in this system could be used for transplantation purpose without of any need to their purification since the cells with which MSCs are co cultured are native cartilage cells. Despite o...

Laminin matrix promotes hepatogenic terminal differentiation of human bone marrow mesenchymal stem cells

Objective(s):The application of stem cells holds great promises in cell transplants. Considering the lack of optimal in vitro model for hepatogenic differentiation, this study was designed to examine the effects of laminin matrix on the improvement of in vitro differentiation of human bone marrow mesenchymal stem cells (hBM-MSC) into the more functional hepatocyte-like cells. Materials and Met...

Bone regeneration with rabbit bone marrow-derived mesenchymal stem cells and bone graft materials.

PURPOSE This study compared the bone regeneration response of different bone graft materials inside canals within anodized titanium implants in cortical and cancellous bone. MATERIALS AND METHODS Upper and lower transverse canals were created in anodic oxidized-surface titanium implants to serve as sites for cortical and cancellous bone regeneration, respectively. The canals were filled with ...
